{"id":11851,"date":"2024-06-11T12:43:38","date_gmt":"2024-06-11T12:43:38","guid":{"rendered":"http:\/\/www.sci.muni.cz\/ofiz\/?p=11851"},"modified":"2024-10-29T10:22:56","modified_gmt":"2024-10-29T10:22:56","slug":"characterization-of-aspergillus-fumigatus-secretome-during-sublethal-infection-of-galleria-mellonella-larvae","status":"publish","type":"post","link":"https:\/\/ueb1.sci.muni.cz\/ofiz\/characterization-of-aspergillus-fumigatus-secretome-during-sublethal-infection-of-galleria-mellonella-larvae\/","title":{"rendered":"Characterization of Aspergillus fumigatus secretome during sublethal infection of Galleria mellonella larvae"},"content":{"rendered":"\n<p><strong>Introduction:<\/strong> The fungal pathogen <em>Aspergillus fumigatus<\/em> can induce prolonged colonization of the lungs of susceptible patients, resulting in conditions such as allergic bronchopulmonary aspergillosis and chronic pulmonary aspergillosis.<\/p>\n\n\n\n<p><strong>Hypothesis:<\/strong> Analysis of the <em>A. fumigatus<\/em> secretome released during sub-lethal infection of <em>G. mellonella<\/em> larvae may give an insight into products released during prolonged human colonisation.<\/p>\n\n\n\n<p><strong>Methodology<\/strong><em>: Galleria mellonella<\/em> larvae were infected with <em>A. fumigatus,<\/em> and the metabolism of host carbohydrate and proteins and production of  fungal virulence factors were analysed. Label-free qualitative proteomic  analysis was performed to identify fungal proteins in larvae at 96 hours post-infection and also to identify changes in the <em>Galleria<\/em> proteome as a result of infection.<\/p>\n\n\n\n<p><strong>Results<\/strong>: Infected larvae demonstrated increasing concentrations of gliotoxin and siderophore and displayed reduced amounts of haemolymph carbohydrate and protein. Fungal proteins (399) were detected by qualitative  proteomic analysis in cell-free haemolymph at 96 hours and could be categorized into seven groups, including virulence (<em>n<\/em> = 25), stress response (<em>n<\/em> = 34), DNA repair and replication (<em>n<\/em> = 39), translation (<em>n<\/em> = 22), metabolism (<em>n<\/em> = 42), released intracellular (<em>n<\/em> = 28) and cellular development and cell cycle (<em>n<\/em>  = 53).
