Na\u0161\u00edm c\u00edlem je pochopit, jak funguje biologick\u00e1 magnetorecepce a identifikovat sign\u00e1ln\u00ed dr\u00e1hy bun\u011b\u010dn\u00fdch syst\u00e9m\u016f, kter\u00e9 jsou citliv\u00e9 na p\u0159irozen\u00e9 geomagnetick\u00e9 pole i \u010dlov\u011bkem generovan\u00e9 technick\u00e9 pole.<\/p>\n\n\n\n<p>V\u00fdzkum je prakticky zam\u011b\u0159en na dopady t\u011bchto pol\u00ed na chov\u00e1n\u00ed, orientaci, pam\u011b\u0165 a cirkadi\u00e1nn\u00ed rytmy bun\u011b\u010dn\u00fdch i vy\u0161\u0161\u00edch syst\u00e9m\u016f. Prim\u00e1rn\u00edmi modelov\u00fdmi organismy jsou z\u00e1stupci z t\u0159\u00eddy hmyzu, ale vyu\u017e\u00edvan\u00e9 jsou tak\u00e9 obratlov\u010d\u00ed modely (nap\u0159. <em>Mus musculus<\/em> nebo <em>Danio rerio<\/em>).<\/p>\n\n\n\n<p>Metodika v\u00fdzkumu je postavena na anal\u00fdze prvk\u016f chov\u00e1n\u00ed model\u016f geneticky podobn\u00fdch lidsk\u00e9mu modelu, a to pod vlivem sv\u011btla a magnetick\u00e9ho pole. Pro tento \u00fa\u010del je laborato\u0159 vybavena unik\u00e1tn\u00edmi elektromagneticky st\u00edn\u011bn\u00fdmi komorami.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">C\u00edle pr\u00e1ce<\/h2>\n\n\n\n<p>V\u00fdzkum je zam\u011b\u0159en p\u0159edev\u0161\u00edm na <strong>potvrzen\u00ed a zkoum\u00e1n\u00ed schopnosti zv\u00ed\u0159at vn\u00edmat magnetick\u00e9 pole Zem\u011b<\/strong>. Kompasov\u00fd smysl byl pops\u00e1n ji\u017e u mnoha druh\u016f \u017eivo\u010dich\u016f, ale jeho podstata a v\u00fdznam st\u00e1le nejsou uspokojiv\u011b objasn\u011bny. Na molekul\u00e1rn\u00ed funkci receptoru a jeho lokalizaci v t\u011ble existuj\u00ed pouze hypot\u00e9zy. Dosavadn\u00ed v\u00fdsledky na\u0161\u00ed laborato\u0159e ukazuj\u00ed, \u017ee hmyz\u00ed kompas by mohl b\u00fdt, podobn\u011b jako je tomu u pt\u00e1k\u016f, spojen se zrakovou drahou. Jestli a jak hmyz magnetick\u00e9 pole skute\u010dn\u011b vid\u00ed, je ot\u00e1zkou, kterou se sna\u017e\u00edme rozlu\u0161tit.<\/p>\n\n\n\n<p>Na z\u00e1klad\u011b behavior\u00e1ln\u00edch a bun\u011b\u010dn\u00fdch odpov\u011bd\u00ed na p\u0159esn\u011b definovan\u00e9 z\u00e1sahy:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li><p><strong>fyzik\u00e1ln\u00ed<\/strong> \u2013 jako jsou orientace a intenzita magnetick\u00e9ho pole, vlivy radiofrekven\u010dn\u00edch a 50Hz pol\u00ed, intenzita a vlnov\u00e1 d\u00e9lka sv\u011btla atd.<\/p><\/li>\n\n\n\n<li><p><strong>biologick\u00e9<\/strong> \u2013 chirurgick\u00e9 z\u00e1sahy, mutace nebo vypnut\u00ed specifick\u00fdch gen\u016f pomoc\u00ed RNA interference (knockdown) \u010di metodou CRISPR-Cas (knockout)<\/p><\/li>\n<\/ul>\n\n\n\n<p>lze <strong>vystopovat molekul\u00e1rn\u00ed princip dosud nezn\u00e1m\u00e9ho smyslu pro zemsk\u00fd magnetismus a jeho lokalizaci v t\u011ble<\/strong>.<\/p>\n\n\n\n<p>Zaj\u00edm\u00e1 n\u00e1s <strong>\u00faloha molekuly Kryptochromu<\/strong> (Cry), jej\u00ed\u017e sign\u00e1ln\u00ed aktivitu po aktivaci sv\u011btlem pravd\u011bpodobn\u011b ovliv\u0148uje magnetick\u00e9 pole Zem\u011b.&nbsp;V\u00fdznam na\u0161eho v\u00fdzkumu p\u0159esahuje probl\u00e9m, jak se zv\u00ed\u0159ata orientuj\u00ed a pom\u00e1h\u00e1 posouvat hranice zn\u00e1m\u00e9ho i ve fotochemii a biofyzice. Ukazuje se tak\u00e9, \u017ee opravdu velmi <strong>slab\u00e1 elektromagnetick\u00e1 pole z&nbsp;oboru radiofrekven\u010dn\u00edho spektra<\/strong> naru\u0161uj\u00ed magnetick\u00fd kompas zv\u00ed\u0159at, co\u017e je nejsp\u00ed\u0161e vysv\u011btliteln\u00e9 pr\u00e1v\u011b pomoc\u00ed zapojen\u00ed Cry. Z\u00e1rove\u0148 je to v\u016fbec nejcitliv\u011bj\u0161\u00ed p\u0159\u00edpad biologick\u00e9 citlivosti k&nbsp;takov\u00fdm vln\u00e1m. Vzhledem k&nbsp;tomu, \u017ee Cry maj\u00ed v\u0161echna zv\u00ed\u0159ata v\u010detn\u011b \u010dlov\u011bka, nab\u00edz\u00ed se ot\u00e1zka, zdali nejsou n\u011bkter\u00e9 na\u0161e funkce vyu\u017e\u00edvaj\u00edc\u00ed Cry (t\u0159eba biorytmy) p\u0159ece jen citliv\u00e9 k&nbsp;radiov\u00fdm vln\u00e1m a jak by se toho p\u0159\u00edpadn\u011b dalo vyu\u017e\u00edt. Mysl\u00edme si, \u017ee v\u0161ude, kde je Cry n\u011bjak zapojen v&nbsp;\u0159\u00edzen\u00ed bun\u011b\u010dn\u00fdch d\u011bj\u016f, nese si magnetickou citlivost s&nbsp;sebou. Proto\u017ee poruchy biologick\u00e9 rytmicity jsou spojeny s&nbsp;mnoha chorobami od onkologick\u00fdch po psychick\u00e9, chceme se o vztahu Cry \u2013 magnet dozv\u011bd\u011bt v\u00edce.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Metody v\u00fdzkumu<\/h2>\n\n\n\n<p>Z\u00e1kladn\u00edm typem experimentu je <strong>sledov\u00e1n\u00ed chov\u00e1n\u00ed zv\u00ed\u0159at nebo i bun\u011b\u010dn\u00e9ho metabolismu<\/strong> za r\u016fzn\u00fdch, p\u0159\u00edsn\u011b hl\u00eddan\u00fdch podm\u00ednek. Dv\u011b elektromagneticky st\u00edn\u011bn\u00e9, unik\u00e1tn\u00ed laborato\u0159e jsou vybaveny velk\u00fdmi c\u00edvkami pro automatick\u00e9 nastaven\u00ed libovoln\u00fdch magnetick\u00fdch parametr\u016f a videosyst\u00e9my (kamera, PC, anal\u00fdza obrazu) pro z\u00e1znam a vyhodnocov\u00e1n\u00ed orienta\u010dn\u00edho chov\u00e1n\u00ed zv\u00ed\u0159at. Vedle toho sledujeme cirkadi\u00e1nn\u00ed rytmy aktivity, kter\u00e9 jsou na Cry z\u00e1visl\u00e9 a rovn\u011b\u017e citliv\u00e9 na magnetick\u00e9 pole.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">\u010clenov\u00e9 laborato\u0159e<\/h2>\n\n\n\n<ul class=\"wp-block-list\">\n<li><a href=\"https:\/\/is.muni.cz\/auth\/osoba\/150822\">Mgr. Spr\u00e1vn\u00fd cirkadi\u00e1nn\u00ed rytmus je p\u0159itom v pozad\u00ed mnoha funkc\u00ed organismu a jeho poruchy vedou k \u0159ad\u011b chorob. Experiment\u00e1ln\u00ed behavior\u00e1ln\u00ed pr\u00e1ce na hmyz\u00edch a sav\u010d\u00edch modelech m\u00e1 uk\u00e1zat, jak\u00e1 je citlivost tohoto jevu a jak\u00fd je jeho mechanismus a podstata.<\/p>\n<\/div><\/div>\n\n\n<div class=\"su-spoiler su-spoiler-style-fancy su-spoiler-icon-chevron-circle su-spoiler-closed\" data-scroll-offset=\"0\" data-anchor-in-url=\"no\"><div class=\"su-spoiler-title\" tabindex=\"0\" role=\"button\"><span class=\"su-spoiler-icon\"><\/span>Kryptochrom jako kandid\u00e1t na magnetosensitivn\u00ed molekulu zv\u00ed\u0159at<\/div><div class=\"su-spoiler-content su-u-clearfix su-u-trim\">\n<p>V\u00edme, \u017ee laboratorn\u00ed hmyz dok\u00e1\u017ee vn\u00edmat sm\u011br magnetick\u00e9ho pole Zem\u011b. Mechanismus kompasu zv\u00ed\u0159at je dosud nezn\u00e1m\u00fdm zp\u016fsobem v\u00e1z\u00e1n na flavoprotein Kryptochrom (Cry). Pomoc\u00ed genov\u00e9 editace (Crispr) a p\u0159edev\u0161\u00edm behavior\u00e1ln\u00edch metod na hmyz\u00edch modelech budeme zkoumat, jak Cry funguje a jestli je opravdu hledan\u00fdm magnetoreceptorem.<\/p>\n<\/div><\/div>\n\n\n<div class=\"su-spoiler su-spoiler-style-fancy su-spoiler-icon-chevron-circle su-spoiler-closed\" data-scroll-offset=\"0\" data-anchor-in-url=\"no\"><div class=\"su-spoiler-title\" tabindex=\"0\" role=\"button\"><span class=\"su-spoiler-icon\"><\/span>Schopnosti u\u010den\u00ed v\u010dely medonosn\u00e9 pod vlivev elektromagnetick\u00fdch pol\u00ed<\/div><div class=\"su-spoiler-content su-u-clearfix su-u-trim\">\n<p>Publikovan\u00fd experiment ukazuj\u00edc\u00ed, \u017ee schopnost v\u010del u\u010dit se je v\u00fdznamn\u011b sn\u00ed\u017eena v bl\u00edzkosti 50Hz elektromagnetick\u00fdch pol\u00ed v okol\u00ed silov\u00fdch veden\u00ed, pot\u0159ebuje nez\u00e1visle ov\u011b\u0159it.
