U obratlovc\u016f se vyskytuje je\u0161t\u011b imunita adaptivn\u00ed, kter\u00e1 se vyv\u00edj\u00ed b\u011bhem \u017eivota jedince. Srovn\u00e1vac\u00ed imunologie studuje imunitn\u00ed reakce \u017eivo\u010dich\u016f b\u011bhem evolu\u010dn\u00edho v\u00fdvoje, hled\u00e1 rozd\u00edly nebo naopak shodn\u00e9 procesy udr\u017een\u00ed st\u00e1l\u00e9ho vnit\u0159n\u00edho prost\u0159ed\u00ed a obrany proti patogen\u016fm. St\u00e1le v\u00edce je kladen d\u016fraz na p\u0159irozenou str\u00e1nku imunity, kter\u00e1 reaguje na patogeny nejrychleji, produkuje antibakteri\u00e1ln\u00ed l\u00e1tky, eliminuje cizorod\u00e9 \u010d\u00e1stice a je sou\u010d\u00e1st\u00ed nap\u0159. koagulace t\u011bln\u00edch tekutin. Studiem imunity bezobratl\u00fdch je mo\u017en\u00e9 pochopit i podobn\u00e9 reakce u mnohem slo\u017eit\u011bj\u0161\u00edch obratlovc\u016f. Metody studia p\u0159irozen\u00e9 imunity jsou velmi podobn\u00e9 u bezobratl\u00fdch i obratlovc\u016f, proto jsme se zam\u011b\u0159ili na studium hmyzu, ryb a pt\u00e1k\u016f (z\u00e1stupce bezobratl\u00fdch, studenokrevn\u00fdch a teplokrevn\u00fdch obratlovc\u016f).<\/p>\n<p><a href=\"http:\/\/www.sci.muni.cz\/ofiz\/wp-content\/uploads\/2019\/06\/vitr\u00edna.jpg\"><img loading=\"lazy\" decoding=\"async\" class=\"wp-image-2294 size-large aligncenter\" src=\"http:\/\/www.sci.muni.cz\/ofiz\/wp-content\/uploads\/2019\/06\/vitr\u00edna-1024x682.jpg\" alt=\"\" width=\"940\" height=\"626\" srcset=\"https:\/\/ueb1.sci.muni.cz\/ofiz\/wp-content\/uploads\/2019\/06\/vitr\u00edna-1024x682.jpg 1024w, https:\/\/ueb1.sci.muni.cz\/ofiz\/wp-content\/uploads\/2019\/06\/vitr\u00edna-300x200.jpg 300w, https:\/\/ueb1.sci.muni.cz\/ofiz\/wp-content\/uploads\/2019\/06\/vitr\u00edna-768x512.jpg 768w\" sizes=\"auto, (max-width: 940px) 100vw, 940px\" \/><\/a><\/p>\n<p style=\"text-align: justify;\">V r\u00e1mci velmi obs\u00e1hl\u00e9 problematiky fyziologie a imunologie bezobratl\u00fdch jsme se zam\u011b\u0159ili na studium obrann\u00fdch reakc\u00ed hmyzu. Hmyz\u00ed imunitn\u00ed syst\u00e9m (slo\u017een\u00fd z humor\u00e1ln\u00ed a bun\u011b\u010dn\u00e9 imunity) je odli\u0161n\u00fd od sav\u010d\u00edho, i kdy\u017e obsahuje n\u011bkter\u00e9 analogick\u00e9 struktury. V mnoha sm\u011brech jde naopak o zcela origin\u00e1ln\u00ed zp\u016fsob obrany organismu proti patogen\u016fm. To, \u017ee se jedn\u00e1 o jin\u00e9 principy ne\u017e u savc\u016f, je d\u00e1no malou velikost\u00ed hmyzu, jednoduchost\u00ed nervov\u00e9 soustavy, pevnou kutikulou na povrchu t\u011bla a dal\u0161\u00edmi vlastnostmi spojen\u00fdmi s morfologi\u00ed, anatomi\u00ed a fyziologi\u00ed hmyzu.<\/p>\n<p style=\"text-align: justify;\">Obrann\u00e9 reakce jsou vyvol\u00e1ny p\u016fsoben\u00edm stresu a n\u00e1sledn\u011b se sleduj\u00ed zm\u011bny nap\u0159. v proteinov\u00e9m spektru hemolymfy, v produkci voln\u00fdch radik\u00e1l\u016f, v aktivit\u011b antibakteri\u00e1ln\u00edch a antioxida\u010dn\u00edch slo\u017eek atd. Jako stresov\u00fdch faktor\u016f je mo\u017eno pou\u017e\u00edt vliv teploty, poran\u011bn\u00ed, injikace patogen\u016f, p\u0159irozen\u00e9 invaze entomopatogenn\u00edch hl\u00edstovek aj. \u0158adu parametr\u016f lze tak\u00e9 ovlivnit p\u016fsoben\u00edm hormon\u016f a chemick\u00fdch l\u00e1tek z \u0159ad insekticid\u016f a polutant\u016f.<\/p>\n<p style=\"text-align: justify;\">Jako modelov\u00e9 organismy pou\u017e\u00edv\u00e1me bource moru\u0161ov\u00e9ho (<em>Bombyx mori L.<\/em>), zav\u00edje\u010de voskov\u00e9ho (<em>Galleria mellonella L.<\/em>), v\u010delu medonosnou (<em>Apis mellifera<\/em>) a octomilku (<em>Drosophila melanogaster<\/em>). V\u011bt\u0161ina experiment\u016f se prov\u00e1d\u00ed s hemolymfou odebranou modelov\u00fdm organism\u016fm. Mezi pou\u017e\u00edvan\u00e9 metody pat\u0159\u00ed polyakrylamidov\u00e1 elektrofor\u00e9za, luminometrie, biochemick\u00e9 sety a imunologick\u00e9 metody jako jsou ELISA, radi\u00e1ln\u00ed dif\u00faze v agar\u00f3ze aj.<\/p>\n<p style=\"text-align: justify;\">V posledn\u00ed dob\u011b je velmi aktu\u00e1ln\u00ed problematika entomopatogenn\u00edch hl\u00edstovek, kter\u00e9 maj\u00ed velk\u00fd v\u00fdznam pro biologickou kontrolu hmyz\u00edch \u0161k\u016fdc\u016f. Pro experimenty pou\u017e\u00edv\u00e1me druhy <em>Heterorhabditis bacteriophora <\/em>a <em>Steinernema feltiae<\/em>. Tyto druhy se li\u0161\u00ed svoj\u00ed patogenitou a tak\u00e9 jsou odli\u0161n\u011b rozpozn\u00e1v\u00e1ny imunitn\u00edm syst\u00e9mem hmyz\u00edho hostitele. Experimenty prob\u00edhaj\u00ed zejm\u00e9na na octomilce <em>Drosophila melanogaster<\/em> a zav\u00edje\u010di voskov\u00e9m <em>Galleria mellonella<\/em> p\u0159i r\u016fzn\u00e9 teplot\u011b a koncentraci hl\u00edstovek.<\/p>\n<p style=\"text-align: justify;\">Entomopatogenn\u00ed hl\u00edstovky rodu <em>Heterorhabditis<\/em> \u017eij\u00ed v symbi\u00f3ze s bakteriemi rodu <em>Photorhabdus<\/em>, kter\u00e9 jsou jedin\u00e9 terestrick\u00e9 p\u0159irozen\u011b bioluminiscen\u010dn\u00ed bakterie. Z\u00e1kladem v\u00fdzkumu v t\u00e9to oblasti je luminometrick\u00e1 detekce bioluminiscence a jej\u00ed ovlivn\u011bn\u00ed optickou hustotou, teplotou atd. Vyu\u017eit\u00ed se nab\u00edz\u00ed v nejr\u016fzn\u011bj\u0161\u00edch detek\u010dn\u00edch syst\u00e9mech zalo\u017een\u00fdch na viabilit\u011b bakteri\u00ed.<\/p>\n<p style=\"text-align: justify;\">Pro studium bakteri\u00e1ln\u00ed infekce hmyzu pou\u017e\u00edv\u00e1me p\u0159irozenou invazi entomopatogenn\u00edch hl\u00edstovek do larev octomilek, kde zablokov\u00e1n\u00edm vybran\u00fdch gen\u016f (mutantn\u00ed nebo RNAi linie) doch\u00e1z\u00ed ke zv\u00fd\u0161en\u00ed citlivosti k n\u00e1kaze hl\u00edstovkami. Touto metodou studujeme zejm\u00e9na zapojen\u00ed koagulace hemolymfy do imunitn\u00edch proces\u016f a tak\u00e9 roli eikosanoid\u016f, jedn\u00e1 se o funk\u010dn\u00ed ov\u011b\u0159en\u00ed kandid\u00e1tn\u00edch gen\u016f.<\/p>\n<p style=\"text-align: justify;\">P\u0159i v\u00fdzkumu imunity v\u010del jsme se zam\u011b\u0159ili na studium jejich dlouhov\u011bkosti, tj. na odli\u0161nost letn\u00ed a zimn\u00ed generace v\u010del. Krom\u011b bun\u011b\u010dn\u00e9 a humor\u00e1ln\u00ed imunity v\u010dely disponuj\u00ed je\u0161t\u011b soci\u00e1ln\u00ed imunitou, kde se zapojuje slo\u017eit\u00e9 chov\u00e1n\u00ed a d\u011blba pr\u00e1ce uvnit\u0159 v\u010del\u00edho spole\u010denstva. M\u011b\u0159en\u00ed fyziologick\u00fdch a imunitn\u00edch parametr\u016f je d\u016fle\u017eit\u00e9 pro stanoven\u00ed z\u00e1t\u011b\u017ee\u00a0 zp\u016fsoben\u00e9 parazity, patogeny, p\u016fsoben\u00ed chemick\u00fdch l\u00e1tek apod.<\/p>\n<p style=\"text-align: justify;\">P\u0159i studiu p\u0159irozen\u00e9 imunity ryb a pt\u00e1k\u016f analyzujeme ve spolupr\u00e1ci s dal\u0161\u00edmi pracovi\u0161ti nap\u0159. fagocyt\u00e1rn\u00ed vlastnosti b\u00edl\u00fdch krvinek (oxida\u010dn\u00ed vzplanut\u00ed) a antibakteri\u00e1ln\u00ed vlastnosti krevn\u00ed plasmy (komplementov\u00fd syst\u00e9m, lysozym).
